Synthesis of Phenylpyridylbutenoid Dimers and their Inhibitory activity against NO Production

Abstract
Several naturally occurring phenylbutenoid dimers were reported to possess anti-inflammatory activity. Three phenylpyridylbutenoid dimers(#1, #2 and #3) were synthesized and evaluated for their inhibitory activities against NO production from LPS-induced RAW 264.7 cells. None of the tested phenylpyridylbutenoid dimers showed any inhibitory activity against NO production, exhibiting cytotoxicity at a concentrated(100 ¥ìM) dose. These results imply that substitution of the benzene ring-A of the natural phenylbutenoid dimer(R=OMe) to a less hydrophobic pyridyl group results in loss of bioactivity.
